Bioelectroremediation of hexadecane in electrical cells containing Aspergillus niger immobilized in alginate

چکیده

Bioelectroremediation (BER) uses electrical current to stimulate catabolism of environmental pollutants, such as petroleum spills. However, applying soil may have adverse effects on the microorganisms involved in degradation. We identify and evaluate BER capabilities a strain Aspergillus niger obtained from petroleum-contaminated soil. Spores this immobilized alginate spheres (2 g) were mixed with 100 g hexadecane-contaminated sandy loam exposed 5, 10, or 15 mA direct 200 cm3 cell copper electrodes. Soil hexadecane concentration was measured by gas chromatography. More than 94% removed within 12 days for currents tested, A. grew 6 x106 CFU g-1 at 10 current. The maximum degradation achieved using 10-mA 20 days, but more 99% fifth day. These results suggest that use spore-containing beads promotes growth biodegradation currents.

Affinity precipitation of Aspergillus niger pectinase by microwave-treated alginate.

Affinity precipitation is a simple, single plate separation process in which the complex of a smart macroaffinity ligand with the target protein (from a crude broth) can be selectively precipitated by application of a suitable stimulus. Alginate is a copolymer of guluronic acid and mannuronic acid residues and precipitates with Ca(2+) ions. It was found to bind to pectinase present in a commerc...

Biodegradation of high concentrations of hexadecane by Aspergillus niger in a solid-state system: kinetic analysis.

Solid-state microcosms were used to assess the influence of constant and variable C/N ratios on the biodegradation efficiency by Aspergillus niger at high hexadecane (HXD) concentrations (180-717 mg g-1). With a constant C/N ratio, 100% biodegradation (33-44% mineralization) was achieved after 15 days, at rates increasing as the HXD concentration increased. Biomass yields (YX/S) remained almost...

Catalytical Properties of Free and Immobilized Aspergillus niger Tannase

A fungal tannase was produced, recovered, and immobilized by entrapment in calcium alginate beads. Catalytical properties of the immobilized enzyme were compared with those of the free one. Tannase was produced intracellularly by the xerophilic fungus Aspergillus niger GH1 in a submerged fermentation system. Enzyme was recovered by cell disruption and the crude extract was partially purified. T...
